目的观察化脓隐秘杆菌自然感染奶牛脾脏组织的病理学变化,并检测组织细胞中Caspase-3、Caspase-8、Caspase-9、Bax和Bcl-2的表达,以确定化脓隐秘杆菌感染对奶牛脾脏组织细胞凋亡的影响。方法筛检2头化脓隐秘杆菌阴性健康奶牛(对照)和5头自然感染化脓隐秘杆菌的奶牛,收集脾脏组织样本,采用本课题组国家发明专利方法,进行石蜡制片和HE染色,观察脾脏组织病理学变化;采用免疫组织化学方法与本课题组国家发明专利方法相结合,检测奶牛脾脏组织中与细胞正负凋亡相关的调节基因Bax和Bcl-2及凋亡通路蛋白Caspase-3、Caspase-8和Caspase-9的表达情况。结果肉眼观察可见化脓隐秘杆菌自然感染奶牛脾脏肿大、柔软,有出血;病理组织学观察可见脾脏组织出血,水肿,淋巴细胞数量减少,有坏死;脾脏淋巴细胞Caspase-3、Caspase-9和Bax表达阳性,与对照组相比,差异有统计学意义(P <0.05),少量淋巴细胞Caspase-8和Bcl-2表达阳性,与对照组相比,差异无统计学意义(P> 0.05)。结论化脓隐秘杆菌能够引起奶牛脾脏淋巴细胞发生坏死和凋亡;可通过调节脾脏中淋巴细胞Bcl-2家族的活性,引起其Bax和Bcl-2比率改变,增加淋巴细胞线粒体内外膜通透性,导致与凋亡相关因子的释放,激活Caspase-9,进一步激活Caspase-3,从而引起脾脏中淋巴细胞发生凋亡。 相似文献

To explore the behavior of radiolytically produced hydrogen release from the waste resin stored in a high integrated container(HIC), and the mechanism of hydrogen diffusion in a near-surface disposal facility, both experimental studies and numerical simulations were performed through an accelerated irradiation test and simulated disposal, respectively. Results indicated that,100 years after disposal, the highest hydrogen concentration appeared in the cell where the HICs were placed. The volume fraction for different scenarios postulated in the numerical simulation was 2.64% for Scenario 1, 2.28% for Scenario 2, and 3.965% for Scenario 3, all of which are lower than the hydrogen explosion limit of 4.1%. Permian marine strata have gradually become a research focus in the world. The marine strata of the Late Permian Dalong Formation (P3d) in the Fenghai area, Fujian Province, have become more and more important as their geochemical characteristics record important geological information and are a good indicator for recovering and reconstructing the paleosedimentary environments and tectonic attributes. The major elements, trace elements and rare earth elements were analyzed by XRF and ICP-MS, respectively. Based on the results of detailed field geological surveys, profile measurements as well as typical sample collection, the tectonic setting and provenance of Permian marine mudstone were comprehensively discussed. The results showed that the Dalong Formation (P3d) was deposited in an active continental margin tectonic environment as revealed by the relation between Fe2O3 + MgO and TiO2 and Al2O3/SiO2. The fingerprint characteristics of Mn, Fe, Co, Ni and REE and the ratio of U/Th, V/Cr, Sr/Ba, (La/Yb)N and V/(V + Ni) indicated that the sedimentary provenance was mainly derived from potassium feldspar, followed by muscovite. Sedimentary water bodies showed a gradually decreasing depositional rate trend, water depth gradually shallowing and paleo-salinity and productivity gradually increasing. Moreover, since transient delamination occurred during sedimentary processes, sedimentary water bodies showed obvious neritic characteristics. It was consistent with the results revealed by lithological and geochemical characteristics. Calcareous mudstone and siltstone transitioned into fine sandstone from bottom to top, indicating paleo-water bodies became shallow. The research results provided good reference and guidance for understanding Permian paleo-sedimentary environments and tectonic attributes of the Yong’an area, Fujian Province, southeastern China. 相似文献

This paper proposes a new filtering method based on the Kalman filtering algorithm for hot-rolled strip flatness measurement system. The system involves processing slowly changing signal, which can be considered as a bounded random process, and its model parameters are completely unknown. The noise rejection strategy in double lasers can generate a compensation signal. Since the initial and accumulative error would lead to negative filter effect or even cause divergence, Kalman filter is integrated to effectively deal with the initial error and enhance convergence. In this setting, the noise rejection strategy is used as a prediction model to constitute a similar Kalman filter. The correlated error caused by measurement error is coped with by a compensation model based on the feature of correlated error to enhance the filter effect. Both theoretical analysis and simulations show that the new algorithm has a better filter effect than the traditional Kalman filtering algorithm for the system. 相似文献

AbstractPilots’ operation has an important effect on flight safety and performance, particularly in the final landing stage when pilots need to deal with complicated operations. This study aims to determine the potential value of flight data and develop a method of evaluating a pilot’s performance during landing phase based on flight quick access recorder (QAR) data from the perspective of risk assessment. First, a Landing Operation Performance Evaluation Model was developed based on risk evaluation principles. Three landing parameters, which are touchdown distance, touchdown vertical acceleration and touchdown pitch angle, were selected as indicators to evaluate the pilots’ landing operation performance in this model. Second, the flight landing operation performance evaluation system (FLOPES) was set up based on the evaluation model. Test results showed that FLOPES can accomplish all calculation flow of operation performance evaluation. Finally, it concluded that this method is a more accurate and effective way for evaluating the landing operation performance of a flight. It could be as a practical tool for airlines to manage landing risk quantitatively and to provide a more practical support for improving training and design in aviation.Practitioner summary: This study aims to determine the potential value of flight data and to develop a method of evaluating pilot’s landing operation performance from the risk evaluation perspective.
